Troxagliptin succinate is used for the treatment of type 2 diabetes. Using DPP-4 taken from Caco-2 cells as the material, the IC50 of troxagliptin was determined to be 5.4nmol/L; and using human, dog, and mouse plasma DPP-4 as the material, the IC50 of troxagliptin was determined IC50 is 4.2nmol/L, 6.2nmol/L and 9.7nmol/L, respectively. The inhibitory ability of troxagliptin on human DPP-4 recombinant protein is better than that of alogliptin and sitagliptin, with IC50 values of 1.3nmol/L, 5.3nmol/L and 16.0nmol/L, respectively. In the enzyme specificity experiment, troxagliptin has extremely low inhibitory ability on DPP-2, DPP-8, DPP-9, prolyl endopeptidase and fibroblast activation protein alpha, and the specificity is greater than 10,000 times. . Further research also found that troxagliptin competitively inhibits DPP-4 and has the characteristics of a slow binding mode. |
